Overview

Zan Yu Dan — the “Special Pill to Aid Fertility” or “Pill that Sponsors Reproduction” — is the classical male-fertility counterpart to Yu Lin Zhu. Both formulas come from Zhang Jingyue’s Jing Yue Quan Shu (1624) and have been the foundation of TCM male-fertility prescribing ever since.

Where Yu Lin Zhu addresses combined Qi, Blood and Kidney deficiency in women, Zan Yu Dan focuses on Kidney Yang and Essence in men — the two substrates that classical TCM holds as essential for sperm production, libido, sexual function and the energetic vitality required for conception. The formula combines warming Yang tonics (Yin Yang Huo, Xian Mao, Ba Ji Tian, Rou Cong Rong, Du Zhong, Lu Jiao Jiao) with Essence-nourishing herbs (Shu Di, Gou Qi Zi, Dang Gui) and digestive support (Bai Zhu, Chen Pi) so that the tonification can be absorbed.

Modern research on Zan Yu Dan and its component herbs — particularly Yin Yang Huo (Epimedium), Ba Ji Tian (Morinda) and Tu Si Zi (Cuscuta) — has shown effects on sperm count, motility and morphology in animal and human studies. Several Chinese hospital trials have used Zan Yu Dan or its modifications as the main intervention for oligospermia and asthenozoospermia.

TCM pattern

Zan Yu Dan is prescribed for Kidney Yang and Essence deficiency in men, with or without Spleen Qi deficiency:

  • Difficulty conceiving with no obvious female factor
  • Low sperm count, poor motility, abnormal morphology
  • Impotence and erectile difficulties
  • Premature ejaculation
  • Low libido
  • Cold body, particularly cold lower abdomen and extremities
  • Sore, weak lower back and knees
  • Fatigue, lassitude, lack of stamina
  • Frequent clear urination, nocturia
  • Pale complexion
  • Tongue — pale, possibly with teethmarks, white coat
  • Pulse — deep, weak, slow at the proximal (Chi) position

  2. Xian Mao (Curculigo orchioides) — warms Ming Men, strengthens Kidney Yang

  7. Lu Jiao Jiao (degelatinated deer antler) — nourishes Essence and tonifies Yang
  8. Shu Di Huang — nourishes Kidney Yin and Essence
  9. Gou Qi Zi — nourishes Liver-Kidney Yin and Essence
  10. Dang Gui — nourishes Blood
  11. Bai Zhu — tonifies Spleen Qi so the formula is well absorbed
  12. Chen Pi — moves Qi and prevents stagnation from the rich tonics

Comparisons with related formulas

  • You Gui Wan — broader Kidney Yang tonification but less focus on sperm and male fertility specifically.
  • Wu Zi Yan Zong Wan — the “Five Seeds” formula, a gentler male fertility formula focused on Kidney Yang and Essence with fewer ingredients (Tu Si Zi, Gou Qi Zi, Wu Wei Zi, Fu Pen Zi, Che Qian Zi). Often combined with Zan Yu Dan or used as a milder alternative.
  • Jin Suo Gu Jing Wan — for Kidney Essence leakage with prominent spermatorrhoea but less broadly tonifying.
  • Er Xian Tang — uses similar Yang and Yin tonics but balanced toward menopausal use in both men and women.
  • Yu Lin Zhu — the female-fertility counterpart from the same source.

Modifications

  • For pronounced low sperm count, combine with Wu Zi Yan Zong Wan
  • For Damp-Heat in the lower burner (prostatitis pattern), modify with Bi Xie, Che Qian Zi and Huang Bai
  • For Liver Qi stagnation with stress-driven sexual dysfunction, combine with Xiao Yao San or Chai Hu Shu Gan San principles
  • For Blood stasis (varicocele), add Tao Ren, Hong Hua and Wang Bu Liu Xing
  • For Spleen Qi deficiency (poor digestion), increase Bai Zhu and add Huang Qi
  • Course of treatment: usually 3 months minimum (one full spermatogenic cycle) to see meaningful change in semen parameters

Cautions

Not appropriate for Yin-deficient Heat patterns — the warming Yang herbs will aggravate hot flushes, night sweats, dry mouth, irritability and red tongue with peeled coat.

Not appropriate during acute illness, fever, infection or active inflammatory conditions.

Use with caution in patients with hypertension, prostate cancer or other hormone-sensitive conditions; always disclose to your medical team.

The formula contains Lu Jiao Jiao (degelatinated deer antler). Our clinic uses only deer antler that meets welfare standards from harvested antler; we do not use any endangered or controlled animal products. Vegetarian patients can request substitution with alternative Yang tonics.
